Do you know High throughput screening?
Introduction
High throughput screening (HTS), is a technology system based on the experimental method of molecular and cellular level, micro-plate form as experimental tool carrier, operating system to perform automated testing process, sensitive and rapid detection equipment to collect experiment data, computer doing the analysis of data and at the same time detecting tens of millions of samples. It has the characteristics of trace, rapid, sensitive and accurate.
Features
High-throughput screening is boring for it has tens of millions of samples to be detected everyday. The operate is prone to feel tired and make mistakes. Automation system consists of a computer and its operating software, automated pipetting devices, incubation centrifugal equipment and stack. Automation system has many advantages over manual operation. It controls the whole process by the use of computer and its software, and the programming process is clear and concise.
Application
Full use of medicinal resources
The high throughput screening depends on a large number of samples, making it possible for the large-scale drug screening. It not only improves the chances of drug discovery but also increased the quality of drug discovery.
Micro screening system
The high throughput screening uses screening models at cellular and molecular level. The sample amount is generally in micrograms (μg), thus saving sample resources as well as experimental material.
Highly automated operation
With the emphasis on high throughput drug screening continues to improve, operation equipment and testing instruments used have made considerable progress. Computer-controlled automation achieved, incidence of operate error reduced and efficiency and accuracy of the results improved.
Combination of multi-disciplinary theories and techniques
In high throughput screening process, not only general pharmacological techniques and theory were applied, but also closely bind to medicinal chemistry, molecular biology, cell biology, mathematics, microbiology, computer science and other subjects.

Simple blood test can tell your age?
In a recently published study in Scientific Reports, scientists from Uppsala University have developed a simple blood test technology that can help reveal the body’s biological age, that is can tell your age.
Professor UIf Gyllensten pointed out that with the popularity of new technologies, the future will easily bring incentive medical interventions or help change the lifestyle of patients and monitor the effect. As the aging of our body and different organs, lifestyle factors such as smoking and stress will affect the body’s molecular processes and the aging rate.
Today, herein researchers have developed a simple blood detection technology that can help reveal how lifestyle affects our body’s aging. The researchers analyzed the level of various proteins in the blood sample form 1,000 individuals. By analyzing the protein levels can help evaluate the body’s biological age, weight, height and hip. Protein expression in the blood can be affected by a series of lifestyles and certain factors will accelerate the aging of the body, while other factors will slow the aging processes.
The researchers said, “We found smoking, high BMI and consumption of sugar-sweetened beverages to increase the predicted chronological age by 2-6 years, while consumption of fatty fish, drinking moderate amounts of coffee and exercising reduced the predicted age by approximately the same amount.” Thus, this method can be applied to dried blood spots and my be useful in forensic medicine to provide basic anthropometrical measures for an individual based on a biological evidence.

Novel Anti-Cancer Method
In a novel study recently published in Nature Structural and Molecular Biology, scientists from the University of Surrey have found some important proteins that perform and regulate key biological process. When those proteins or the process fail to function, cancer and other diseases would come. This finding may help develop more effective methods for the treatment of incurable and debilitating disease.
In this study, the research team has, for the first time, analyzed a kind of important regulatory proteins-RNA binding proteins in baker’s yeast and roundworms as well as their interactions with mRNA (an important molecule that guides protein synthesis). Although these two organisms are very different, but scientists discovered in the protein revealed amazing evolutionarily conservation, which means however, metabolic enzymes-the team believes it has existed for billions of years-have new features.
André Gerber, first author of this article said: “This is an important breakthrough, because the protein and process we identified also present in human beings. We knew that they have a strong relationship with a number of diseases like cancers and neurodegenerative diseases. Now we can use new knowledge to test if the case-a new mechanism that control key biological processes-can bring effective treatment for cancers.”
Dr. Emma Laing, co-author of this article said: “It may be that before the show of protein, RNA drives this function, going back to the old mechanism, or it may be that these proteins have a hidden feature that we do not know now.”

Organoid in Laboratory: “Kidney”
In a report recently published in Nature, scientists from Australia and the Netherlands said they have successfully cultured human kidney tissue that has a preliminary growth state by using stem cells, making the idea of developing fully functional organ in the laboratory one step further!
The researchers said this tissue is not viable but can be used for other purposes, such as replacing animal model in drug toxicity tests. By the application of iPS, researchers produced the kidney-like structure. At present, for the lack of organ donation, using stem cells to create human organs has long been a major obstacle in scientific research.
Scientists need to induce stem cells so that they can differentiate and assemble as kidney, liver and lung cells. Then these cells can form as a real anatomical structure and finally function in patients. In this very study, the researchers managed to induce pluripotent stem cells to be transformed into two types of adult cells. David, one of the researchers, said: this product is not kidney but a kind of organoid. So there’s still a long way to go for it to be transplanted organ in clinical use.
However, the researchers pointed out that this type of organ might help complete a different medical needs, namely the detection of new drug safety in human body. Stem cells are a kind of primitive cells and after growth and differentiation, they are developed into a variety of special types of cells which will form different organs such as brain, heart, kidneys and so on.
Until a few years ago, when the iPS has been developed, the only way to get stem cells is from human embryos. However, the destruction of human embryos for the obtain of stem cells is controversial in the scientific community. Currently, other research teams have also reported to have cultured organoids like stomach, liver, retina, brain and heart tissue by the use of pluripotent stem cells.

Microbial Cloud around You
We leave hints of bacteria on all that we touch-keyboard, cell phone, water cups, however, physical contact isn’t the only way to make a microbial mark. People shed swarms of microscopic organisms that linger in the space around them, making a “microbial cloud”.
Published in Peer J, the new study revealed that such cloud of bacteria may be totally unique to each person., just like a microbial fingerprint. Before came to this conclusion, scientists have conducted two experiments in which they placed people in test in a sanitized room and surrounded them with rings of petri dishes. To catch the floating microbes, they suspended air filters. As comparison, another room is humanless but otherwise identical. They compared the samples captured from the two rooms and found that the two species were microbially distinct. Then, another question came into being: if a person’s microbial can set two rooms apart, can it set two people apart? The answer was yes in six of the participants.
Furthermore, the researchers sequenced the microbial cloud samples and found that many of the participants have the same bacteria in their cloud, but in different concentrations. As for how far our microbial cloud can extend, the researchers were not sure. But they estimated that the cloud float within a 90 centimeter radius, suggesting that there’s a high chance that our microbial cloud will mingle in any semicrowded space like in a subway.
So , can you imagine that, a microbial cloud around you?

How does fish beat the heat?
First think about it: when we feel too hot, what will we do to beat the heat? Air conditioning, fan, swimming in the pool, etc. would be the things that come into mind. Yes, we beat heat with those things. But what will fishes do, if they feel hot when the temperature of the water is so hot? A recent new study has given an answer to it.
A team in James Cook University has found that in order to withstand a warming climate, fishes would retreat to the deeper ocean floor. This study, published in Coral Reefs however, provided direct evidence for the impact of climate change bringing to ocean ecology.
Researchers have labeled 60 Redthroat Emperor Fish and released them at Heron Island, which is in the south of the Great Barrier Reef in east Australia. These fish are equipped with an electronic signaling device that can send a signal, it can provide the number, depth, and location information to the receiver. When these fish appear in the Great Barrier reef slopes, signals emitted by those electronic devices can be monitored. By year-long monitoring, the researchers found that these fish appear more in the reef slope when the temperature is low. At the same time, they found that when the temperature becomes higher, frequency of these fish appearing in deeper ocean increases.
The researchers considered the effects of temperature, barometric pressure, rainfall, wind and lunar gravity on the positioning of these fish. However, they found only the temperature was related to the occurrence of signals. By one year's monitoring, they found only when the temperature is below 24 degrees Celsius, these signals will be emerge. The researchers believe that there are other options for these fish. On the one hand to escape the heat, they can dive into the deep waters for these fish can tolerate a 160-meter-deep water environment. In addition, they can move farther, and migrate to the south ocean where the water temperature is lower. Because the researchers found the whereabouts of these fish in the west coast of Australia Perth coast, far away from the habitat of these fish.
The next step of them is to focus on whether these fish can adapt to high temperature environment, Leanne Currey, one of the researchers in this study, commented.
